Abstract: Medicinal plants are regarded as rich resources of traditional medicines and from these plants many of the modern medicines are produced. So this study was designed to evaluate the phytochemical composition of methanol extract of Citrus volkameriana Ten. & Pasq. (C. volkameriana Ten. & Pasq) and assesse the safety and some biological activities as anti-inflammatory, anti-nociceptive, anti-ulcer and hepato-protective activity and also determination of in vivo and in vitro antioxidant activity of extract. The results of this study proved that C. volkameriana Ten. & Pasq leaves methanol extract is non toxic to mice up to 5 g/kg b.wt., and exhibited significant anti-inflammatory activity at 1000 mg/kg b.wt. in rats, anti-nociceptive activity at 500& 1000 mg/kg b.wt, anti-ulcer and hepatoprotective in all dose used. Also, the extract showed potent in vivo antioxidant activity appeared by significant increase in liver GSH, decrease liver MDA level and increase liver NO level in three doses as compared to paracetamol group. Similarly in- vitro affirmed the antioxidant activity of extract by using 1,1- a diphenyl 2-picryl hydrazyl (DPPH) radical assay. Phytochemical evaluation proved the presence of flavonoids, tannins, and triterpenes.
