Analytical method development and validation for estimation of indapamide in bulk drug and dosage form by UV-Spectroscopy

Abstract: Background: A novel and reliable UV spectrophotometric method has been established and verified for quantifying indapamide in both bulk drug and pharmaceutical tablet formulations. This approach utilizes direct UV spectrophotometry for precise analysis.Materials and Methods: Spiked indapamide solution was scanned over UV-visible range for its wavelength of maximum absorbance. Various calibration standards of indapamide were prepared, and absorbance was recorded at wavelength of maximum absorbance. Calibration curve of concentration vs. absorbance was plotted and linearity and range was calculated. Various analytical method validation parameters viz. accuracy, precision, LOD, LOQ, robustness and ruggedness were calculated using QC standards. In order to check the performance, pre-validated method was used to estimate the indapamide content of marketed formulation.Results and Discussion: The sample is dissolved in a suitable solvent, such as water: methanol, and subjected to analysis using a UV-Vis spectrophotometer. Detection is performed at a wavelength specifically optimized for indapamide, typically around 241 nm to achieve optimal sensitivity. The method's validation includes assessing parameters such as linearity, accuracy, precision (both repeatability and intermediate precision), limit of detection, limit of quantification, and robustness. Conclusion: This straightforward and accurate UV spectrophotometric method enables efficient and routine analysis of indapamide in bulk drug and tablet formulations. It offers a cost-effective approach that can be readily implemented in pharmaceutical quality control laboratories for consistent monitoring and testing of indapamide.
