Adverse effects associated with proton pump inhibitors

Abstract: The introduction of PPI to market in from 1989 which is used for management of gastric acid related disorder. Due to normality, the potential mechanism of PPI in blocking the gastric acid secretion by parietal cells with few tolerable side effects has quickly displaced. Other compounds like H2 antagonist and the drug of choice for peptic ulcer, Zollinger-Ellison syndrome, and non-steroidal anti-inflammatory drug associated ulcer, gastro esophageal ulcer, and eradication of helicobacter pylori. This PPI has exponential increase in the prescriptions in recent years. The widespread of PPI has also emerged to long term adverse effects like risk of kidney, liver, dementia and cardiovascular disease, enter endocrine tumor, respiratory and gastro intestinal infection and impaired absorption of drugs and nutrients. All the published evidence has no strong correlation with safety of PPI and reconsideration of clinical indications. Hence, the aim of the review is to evaluate the association of PPI and its serious adverse effects given by overcome of PPI in general population.
